Microscope PhotometerOptical Adjustement
Centre the low-power light source
insert the diffusion disc
Centre the photometer light source
Insert the diffusion disc (unnecessary with fluorometry)
Set up the adjustment sample at low to medium magnific-
ation (low-power illumination)
For measurements in polarized light only:
Centre the objectives. set the polarizer
Swing out the field lens
Only when the grating monochromator is used:
Insert the 0.5mm pair of slits
Set the stray light filter turret (390-80onm)
Centre and focus the field diaphragm of the photometer
Adjust the monochromator slit
With reflection measurements also set the aperture dia-
phragm and half stop.
Insert the pair of slits for the band width required.
Place the sample on the object stage. select the desired
objective. for transmitted light also select the condenser
objective and. if necessary. set the aperture diaphragm.
Disengage the short-circuit lens.
Centre the measuring diaphragm.
Adjust the measuring diaphragm illumination (pilot light).
Determine the size of the measuring field.
Adjust the field of Illumination to the measuring field.
Set the desired spectral region by means of the mono-
chromator or filter.
If necessary refocus object and field diaphragm.
